蓋盒呈圓形，蓋與盒身子母口相合，通體施白釉，釉色潔白細膩，以五彩繪穿花龍紋裝飾。蓋面紅、藍雙龍，龍鱗清晰，首尾相對，身姿虯曲彎繞，四周填以纏枝花紋，黃、綠纏枝線條婉轉流暢，紅黃花盛開繽紛。盒蓋側面及盒身外壁均繪四條龍紋，盒蓋側面為紅、藍、綠龍，呈追逐寶珠之姿，飄逸靈動，極具神采，周圍襯以纏枝花紋。底無釉露胎，中央圓形開光施白釉，青花「大明萬曆年製」雙行六字楷書款。
萬曆五彩燒造極為複雜，傳世稀少。工匠在燒造五彩瓷器時探索出諸多彩料呈色的最佳溫度，一改前朝彩瓷兩次燒成的技術，改分四次燒造，終燒出色彩紛呈的萬曆五彩。當時宮廷需求浩繁，宮中有旨：「龍鳳花草各肖形容，五彩玲瓏務極華麗。」萬曆五彩以繽紛華麗稱雄，紅綠的鮮豔搭配是為向世人展現皇室生活的奢華。以五彩繪龍紋圖蓋盒之例可見收錄於《中國名陶百選》明萬曆〈五彩龍鳳紋蓋物〉，可資參考。細觀此器，胎體敦實，青花色調深沉藍豔，紅彩蒼雅深沉，綠彩翠意閃現，諸彩相配得當，相互輝映，構圖層次繁而不亂，畫風自然流暢，為萬曆朝之彩瓷佳品。
